Unlock Dermal Rejuvenation with Palmitoyl Tripeptide-5

Palmitoyl Tripeptide-5 (Pal-Lys-Val-Lys) is a highly penetrating biomimetic lipopeptide that serves as a foundational active for structural skin rejuvenation. Cosmetically positioned as a dual-action collagen booster, the molecule mimics the endogenous extracellular matrix protein thrombospondin-1 (TSP-1) to activate latent transforming growth factor-β (TGF-β). This activation signals dermal fibroblasts to synthesize types I and III collagen, while concurrently inhibiting matrix metalloproteinases (MMP1 and MMP3) to shield the existing dermal matrix from enzymatic degradation.

Market context: — The B2B landscape for cosmetic peptides features prominent branded offerings like Syn®-Coll (from DSM) and Matrixyl (from Sederma, which uses Palmitoyl Tripeptide-1 as a signal peptide for 'retinol-like' efficacy). Formulate Labs exclusively provides the pure INCI, Palmitoyl Tripeptide-5, ensuring formulators access the precise biomimetic amino-acid sequence and proven bioactive performance without the overhead of branded SKU licensing and associated markups.

Our standard Palmitoyl Tripeptide-5 suspension is meticulously formulated in a hydrating glycerin/water base, designed for optimal dermal delivery and integration into a wide array of cosmetic formulations. This approach ensures formulators receive a high-purity, bioavailable active, enabling the full spectrum of its collagen-boosting and protective benefits, aligning with industry best practices for peptide stability and efficacy.

We supply it at 0.1% standard or 1% lab concentrate in glycerin or Sea Kelp Bioferment carriers.

Why formulators reach for it

A peptide with a long cosmetic-science track record.

Palmitoyl Tripeptide-5 Activates Collagen Synthesis
Mimics thrombospondin-1 to activate latent TGF-β, signaling dermal fibroblasts to synthesize essential collagen types I and III, crucial for skin's structural integrity.
Palmitoyl Tripeptide-5 Protects Dermal Matrix
Inhibits matrix metalloproteinases (MMP1 and MMP3), safeguarding existing collagen and elastin from enzymatic degradation and preserving skin's youthful framework.
Palmitoyl Tripeptide-5 for Visible Rejuvenation
Clinical studies demonstrate that topical formulations containing Palmitoyl Tripeptide-5 provide rapid and sustained improvements in the appearance of periorbital and perioral wrinkles.
Palmitoyl Tripeptide-5 Enhances Skin Structure
Supports the overall extracellular matrix, contributing to improved skin volume, density, and structural integrity for a firmer, more resilient complexion.

Formulate Labs supplies the pure INCI, Palmitoyl Tripeptide-5, and does not sell branded ingredients such as Syn®-Coll or Matrixyl.

Published research

Peer-reviewed literature for due diligence.

Palmitoyl Tripeptide-5 (Syn-Coll) is documented as a TGF-beta-activating cosmetic peptide that stimulates procollagen synthesis and supports matrix protection in published peptide reviews. These are independent, third-party papers we link for formulators evaluating mechanism, tolerability, and published panel data. We did not conduct this research and do not guarantee replication in your finished formula.

Compliance & safety

The fine print, in plain English.

What it’s for
Cosmetic raw material - leave-on hair-care formulations (scalp serums, hair tonics, leave-on hair lotions, density-positioned conditioning treatments) at the use rates above; also acceptable in rinse-off cosmetic products and skincare formats where a peptide-based "supports the look of fuller hair" / scalp-comfort positioning is intended. Not a finished consumer product; not for ingestion, injection, or unsupervised topical use outside a formulated finished product.
Claims guidance
Cosmetic structure / function language only (e.g. supports a firmer-looking finish, softens the look of fine lines). Avoid TGF-beta drug language (“activates TGF-beta”, “stimulates collagen production” as a drug claim). Do not make therapeutic claims. The peptide is studied in cosmetic in-vitro and panel work; downstream finished-product claims must be substantiated by the brand.
Storage
  • Store between 2-25 °C in original sealed container away from direct light.
  • Cold storage extends shelf life by ~50% relative to room temperature.
  • Once opened, use within the period stated on the COA.
Regulatory
  • INCI: Palmitoyl Tripeptide-5 (per ICID and PCPC dictionary).
  • Not classified as hazardous based on currently available information per OSHA HazCom 2012 / GHS.
  • Compatible with FDA cosmetic regulations and EU 1223/2009 for the intended use.
  • SDS section 16 disclaimer applies; see the per-recipe SDS for the full classification.
Sensitivity
Palmitoyl Tripeptide-5 is generally well-tolerated in cosmetic use at the rates above; the supplier’s HET-CAM and AMES studies on the peptide indicate non-irritating and non-mutagenic behavior at industry-standard concentrations. The peptide is light-sensitive: store and ship in opaque containers, and avoid prolonged exposure to direct sunlight in finished formulations. Avoid prolonged contact with strong oxidisers, sulfuric or phosphoric acids, acetic anhydride, sustained heat above 45 °C, and prolonged exposure to pH > 7.0 - any of these can hydrolyze the peptide and reduce in-formula activity. Patch-test finished formulations per standard cosmetic protocol.
