The peptide

Palmitoyl Hexapeptide-12: The Advanced Lipopeptide for Dermal Remodeling and Barrier Restoration

Palmitoyl Hexapeptide-12 is a synthetically engineered lipopeptide, strategically designed by combining the elastin-derived "spring fragment" (Val-Gly-Val-Ala-Pro-Gly) with a palmitic acid moiety. This unique structure significantly enhances its stratum corneum penetration and overall bioavailability within the skin [cite: 1, 2]. Positioned in the cosmetic sector as a high-performance firming, restructuring, and barrier-repair active, Palmitoyl Hexapeptide-12 functions primarily as a matrikine signal peptide [cite: 3, 4]. By down-regulating the pro-inflammatory cytokine IL-6, it actively mitigates extracellular matrix degradation while simultaneously stimulating dermal fibroblasts to synthesize new collagen, elastin, fibronectin, and glycosaminoglycans. This makes the peptide highly effective for anti-aging, photo-damage repair, and long-term skin barrier fortification [cite: 3, 7].

Market context: Palmitoyl Hexapeptide-12 is a key active found in premium anti-aging and barrier-repair formulations, including well-known competitor products like Biopeptide EL™ and Dermaxyl™ (both from Sederma/Croda), and SpecPed® POP (Spec-Chem). These competitor offerings are marketed for restoring dermal elasticity, tone, cellular communication, and providing intensive hydration with barrier replenishment. Formulate Labs provides the generic Palmitoyl Hexapeptide-12 INCI material, not these aforementioned proprietary trade-name SKUs.

Our Palmitoyl Hexapeptide-12 is supplied as a standard 0.1% glycerin suspension, designed for seamless integration into your cosmetic formulations while ensuring optimal stability and bioavailability. This ready-to-use format simplifies compounding, allowing formulators to easily achieve desired active concentrations for high-performance skincare applications.

We supply it as a finished aqueous suspension in two carriers - a 35% glycerin / water base for maximum formulation flexibility, or dosed directly into our single-origin Sea Kelp Bioferment. Both ship at the supplier reference of 0.01% or our 10X lab concentrate at 0.1%.

Why formulators reach for it

A peptide with a long cosmetic-science track record.

Palmitoyl Hexapeptide-12 — Dermal Remodeling
Functions as a matrikine signal peptide, stimulating dermal fibroblasts to synthesize new collagen, elastin, fibronectin, and glycosaminoglycans for improved skin structure.
Palmitoyl Hexapeptide-12 — Barrier Fortification
Enhances stratum corneum penetration and co-assembles with native extracellular matrix molecules to structurally repair and fortify the skin’s natural barrier.
Palmitoyl Hexapeptide-12 — Anti-Aging & Firming
Mitigates extracellular matrix degradation by down-regulating pro-inflammatory cytokines, leading to a visible firming and anti-aging effect.
Palmitoyl Hexapeptide-12 — Photo-Damage Protection
Suppresses UVB-induced EGFR and ERK activation, effectively inhibiting photoaging and the expression of matrix metalloproteinase-1 (MMP-1) in dermal fibroblasts.

Formulate Labs supplies the generic Palmitoyl Hexapeptide-12 INCI material only, not the aforementioned proprietary trade-name SKUs.

pH compatibility
Optimal structural stability is maintained strictly between pH 4.5 and 6.5.
Add at
Thermally sensitive; must be incorporated during the cool-down phase (below 40°C).
Mixing
Standard mixing during the cool-down phase (below 40°C) is suitable for incorporation.
Avoid
Degrades rapidly below pH 4.0 and is structurally incompatible with high-concentration AHAs or strong chemical exfoliants.

Compliance & safety

The fine print, in plain English.

What it’s for
Cosmetic raw material - leave-on hair-care formulations (scalp serums, hair tonics, leave-on hair lotions, density-positioned conditioning treatments) at the use rates above; also acceptable in rinse-off cosmetic products and skincare formats where a peptide-based "supports the look of fuller hair" / scalp-comfort positioning is intended. Not a finished consumer product; not for ingestion, injection, or unsupervised topical use outside a formulated finished product.
Claims guidance
Cosmetic structure / function language only (e.g. supports the appearance of fuller-looking hair, supports a smoother-looking finish). Avoid hair-growth drug language (“regrows hair”, “treats alopecia”). Do not make therapeutic claims. The peptide is studied in cosmetic in-vitro and panel work; downstream finished-product claims must be substantiated by the brand.
Storage
  • Store between 2-25 °C in original sealed container away from direct light.
  • Cold storage extends shelf life by ~50% relative to room temperature.
  • Once opened, use within the period stated on the COA.
Regulatory
  • INCI: Palmitoyl Hexapeptide-12 (per ICID and PCPC dictionary).
  • Not classified as hazardous based on currently available information per OSHA HazCom 2012 / GHS.
  • Compatible with FDA cosmetic regulations and EU 1223/2009 for the intended use.
  • SDS section 16 disclaimer applies; see the per-recipe SDS for the full classification.
Sensitivity
Palmitoyl Hexapeptide-12 is generally well-tolerated in cosmetic use at the rates above; the supplier’s HET-CAM and AMES studies on the peptide indicate non-irritating and non-mutagenic behavior at industry-standard concentrations. The peptide is light-sensitive: store and ship in opaque containers, and avoid prolonged exposure to direct sunlight in finished formulations. Avoid prolonged contact with strong oxidisers, sulfuric or phosphoric acids, acetic anhydride, sustained heat above 45 °C, and prolonged exposure to pH > 7.0 - any of these can hydrolyze the peptide and reduce in-formula activity. Patch-test finished formulations per standard cosmetic protocol.
