The peptide

Acetyl Tetrapeptide-3: Reinforcing Hair & Skin Matrix for Enhanced Resilience

Acetyl Tetrapeptide-3 (Ac-KGHK) is a biomimetic signal peptide engineered to mimic a natural extracellular matrix (ECM) protein fragment, designed for cosmetic applications aimed at structural reinforcement. It strategically stimulates the synthesis of crucial ECM proteins, notably laminin and collagens III and VII, to strengthen hair follicle anchoring and fortify the dermal-epidermal junction, thereby supporting enhanced hair resilience and skin integrity.

Market context: While Acetyl Tetrapeptide-3 is prominently featured in trademarked complexes like Capixyl™ (a trade name blending the peptide with *Trifolium pratense* extract, which Formulate Labs does not sell), Formulate Labs exclusively supplies the standalone INCI. This approach offers formulators maximum flexibility and cost-efficiency, freeing them from proprietary blend restrictions and enabling tailored product development.

Formulate Labs typically provides Acetyl Tetrapeptide-3 in a convenient 0.1% standard glycerin suspension, ideal for seamless incorporation into a wide range of cosmetic formulations. This pre-solubilized liquid blend offers formulation flexibility and simplifies handling, allowing chemists to achieve precise active concentrations with ease.

We supply the peptide on its own (without the clover extract) as a finished aqueous suspension in two carriers - a 35% glycerin / water base for maximum formulation flexibility, or dosed directly into our single-origin Sea Kelp Bioferment for formulators who want to ship a pre-built hydrating active in one drop. Both ship at the CapixylTM-equivalent peptide reference of 0.035%, or our 10X lab concentrate at 0.35%.

Why formulators reach for it

A peptide with a long cosmetic-science track record.

Supports Follicular Anchoring
Acetyl Tetrapeptide-3 strengthens the hair follicle's connection to the scalp by stimulating laminin and collagen synthesis.
Extends Hair Growth Cycle
Helps prolong the anagen (growth) phase of hair, promoting healthier, longer-looking hair.
Fortifies Dermal-Epidermal Junction
Enhances the structural integrity of the skin’s vital supportive layers for improved resilience.
Stimulates Key ECM Proteins
Promotes the synthesis of essential extracellular matrix proteins, including collagen III, collagen VII, and laminin.

Acetyl Tetrapeptide-3 refers to the pure INCI ingredient, distinct from trademarked blends like Capixyl™, which Formulate Labs does not supply.

Published research

Peer-reviewed literature for due diligence.

Acetyl Tetrapeptide-3 (the Capixyl peptide sequence) is documented in peer-reviewed hair-science literature for follicle anchoring, extracellular-matrix support, and anagen/telogen balance. These are independent, third-party papers we link for formulators evaluating mechanism, tolerability, and published panel data. We did not conduct this research and do not guarantee replication in your finished formula.

Compliance & safety

The fine print, in plain English.

What it’s for
Cosmetic raw material - leave-on hair-care formulations (scalp serums, hair tonics, leave-on hair lotions, density-positioned conditioning treatments) at the use rates above; also acceptable in rinse-off cosmetic products and skincare formats where a peptide-based "supports the look of fuller hair" / scalp-comfort positioning is intended. Not a finished consumer product; not for ingestion, injection, or unsupervised topical use outside a formulated finished product.
Claims guidance
Cosmetic structure / function language only (e.g. supports the appearance of fuller-looking hair, supports a healthy-looking scalp, helps maintain the look of hair density). Avoid hair-loss drug language ("treats hair loss", "stops hair loss", "regrows hair", "treats alopecia", "hair-regrowth treatment") - these are drug claims and are forbidden on cosmetic products in every market we ship into. Avoid mechanism / pharmacological language ("5-α-reductase inhibitor", "blocks DHT", "DHT modulation", "follicle activator", "inhibits 5-AR") - even when the supplier brochure references these mechanisms in cosmetic in-vitro studies, the in-formula language for a finished product must stay cosmetic. Do not make therapeutic claims (treating, curing, healing). The peptide is studied in cosmetic in-vitro and panel work; downstream finished-product claims must be substantiated by the brand.
Storage
  • Store between 2-25 °C in original sealed container away from direct light.
  • Cold storage extends shelf life by ~50% relative to room temperature.
  • Once opened, use within the period stated on the COA.
Regulatory
  • INCI: Acetyl Tetrapeptide-3 (per ICID and PCPC dictionary).
  • Not classified as hazardous based on currently available information per OSHA HazCom 2012 / GHS.
  • Compatible with FDA cosmetic regulations and EU 1223/2009 for the intended use.
  • SDS section 16 disclaimer applies; see the per-recipe SDS for the full classification.
Sensitivity
Acetyl Tetrapeptide-3 is generally well-tolerated in cosmetic use at the rates above; the supplier’s HET-CAM and AMES studies on the peptide indicate non-irritating and non-mutagenic behavior at industry-standard concentrations. The peptide is light-sensitive: store and ship in opaque containers, and avoid prolonged exposure to direct sunlight in finished formulations. Avoid prolonged contact with strong oxidisers, sulfuric or phosphoric acids, acetic anhydride, sustained heat above 45 °C, and prolonged exposure to pH > 7.0 - any of these can hydrolyze the peptide and reduce in-formula activity. Patch-test finished formulations per standard cosmetic protocol.
